The study assessed the effect of a microbial 6-phytase on amino acids digestibility and its interaction with the lysine content in caecectomised laying hens fed low"=P, maize"=soybean based diets. Effects on the utilisation of N, P, and Ca were also studied in a 2\,$\times$\,2"=factorial arrangement with 2 dietary lysine levels (adequate, ALys and reduced, LLys) either without (Phy-) or with (Phy+) phytase supplementation. The laying hens aged 28 weeks were kept in balance crates and 7 individual hens were allocated to each treatment. Hens were fed the treatment diets for 10 days, and excreta were quantitatively collected during the last 5 days for analysis. Result showed that hens fed on adequate lysine"=based diets with or without phytase supplementation had significantly ($p < 0.05$) higher dry matter intake (DMI) than those fed on marginally low lysine"=based diets with or without phytase supplementation. Phytase addition led to numerical improvement in the N-utilisation but this was not significantly ($p > 0.05$) affected by either lysine or phytase levels. Lysine levels did not significantly ($p > 0.05$) affected the amino acids measured. However, addition of phytase led to numerical improvement of the entire amino acids digestibility (except alanine) by between 2 and 26 units. The phytase\,$\times$\,lysine interaction was only significant ($p < 0.001$) for lysine. Marginal reduction in the dietary lysine level led to significant ($p < 0.05$) reduction in the intake and retention of P. However, phytase addition led to a significant ($p < 0.05$) increase in P retention and utilisation with hens fed on marginally low lysine"=based diets having consistently lower values. While Ca intake was significantly ($p < 0.05$) affected by the dietary lysine level, effect of phytase on Ca utilisation was not significant. In the two test diets, addition of phytase led to improvement in Ca utilisation by between 3.2 and 7.3 units. This study shows that while phytase addition led to significant improvement in P utilisation, there was no corresponding significant improvement in amino acids digestibility except lysine in laying hens when fed marginally low lysine diets.}
